Kwangwoo's research paper was reprinted in the March issue of Annals of the Rheumatic Diseases (IF=12.350; online-published on Sep 28, 2018). Kwangwoo investigated the validity/robustness of Rothman's additive interaction model in assessing gene-gene interactions by simulating genome-wide association study (GWAS) datasets with actual human genetic architecture. This paper reports that Rothman’s additive interaction model induces high false positive rates for interactions between genetic variants with large effect sizes on a disease. The results explain all previous reports about significant interactions between large-effect variants such as the HLA-DRB1 shared epitope allele and the PTPN22 variant in rheumatoid arthritis. The paper will help correct the conclusions about gene-gene interactions from Rothmans’ interaction model in published papers and future researches.
